BioSci Seminar: Architecture of Protein Complexes In Situ by Structural Proteomics

Abstract: Understanding the architecture of protein complexes in their native environment is essential for understanding their biological function. However, many transient and low-affinity interactions are missed by protein-protein interaction mapping methods. We are pioneering novel crosslinking mass spectrometry methods to map weak and transient protein-protein interactions directly in cells, which can lead directly to structure solution by Cryo-EM. Using these approaches, we uncover novel complexes in Salmonella Typhimurium, including previously uncharacterized RNA polymerase interactors. In parallel, we develop a targeted enrichment strategy called PhIX-MS to define the interactomes of human protein assemblies. Applying this approach to the proteasome, we identify and validate the binding sites of TXNL1, UBE3C, and the assembly chaperone PSMD5/S5b. Our methods provide a broadly applicable strategy for dissecting protein complex architecture in native cellular contexts.
